Different people/performers use different techniques for prepartion.
They usually use whatever techniques best suit their individual personal needs.

Proper prep for performing an anal scene can sometimes start out days beforehand.
The performer may decide to watch what they ingest/eat...and some often put themselves
on a strict liquids only diet.

Well, personally I can't tell about girls doing drugs. I've never seen that some performer or director hands out drugs to the girls to ease the pain of anal. Actually, if it's done right it shouldn't hurt anyway. But of course you can never know what the girls take to the bathroom in their bags.

About the preparation... Scenes I watched were usually shot between early and late afternoon. The girls didn't eat that day, if they were about to shoot anal. Then they used an enema, this differs between the studios. Some studios insist on using the enema 2+ times, not just once, to be sure to have a really good result and to minimize the risks of bad surprises during shooting.
